Background and Objective: Sepsis is a life-threatening condition characterized by systemic inflammation and multi-organ damage, with the heart and lungs being particularly vulnerable. Diabetes exacerbates sepsis-induced oxidative stress and tissue injury, highlighting the need for effective adjunctive therapies such as ozone. This study evaluated the effects of ozone therapy as an adjuvant treatment on cardiac and pulmonary tissues in a diabetic mouse model of cecal perforation-induced sepsis. Materials and Methods: A total of 46 Swiss albino mice were used, with diabetes induced in 40 mice using streptozotocin (125 mg/kg, IP). After one month, mice were divided into control and experimental groups, including diabetes, ozone-treated, sepsis-induced, and combined treatment groups. Ozone (20 μg/mL, 1 mL, IP) was administered before or after Cecal Ligation and Perforation (CLP). After 24 hrs, heart and lung tissues were collected for histopathological and biochemical analyses. Results: Sepsis and diabetes significantly increased inflammatory and oxidative damage in cardiac and pulmonary tissues, as evidenced by elevated MDA levels and reduced CAT activity. Ozone therapy reduced tissue inflammation, decreased MDA levels, and increased CAT activity. Pre-treatment with ozone showed greater protective effects than post-treatment. Conclusion: Ozone therapy mitigates cardiac and pulmonary damage in diabetic sepsis, with pre-treatment providing more effective protection than post-treatment.
